The key aspects of these methods and their growing success and future challenges include: the use of genetic variation and/or drug perturbation combined with transcriptional or other molecular profiling changes and clinical outcomes to go beyond correlation to extract causal interactions the use of large-scale supercomputing involving thousands or tens of thousands of processors to score billions of possible network frag- ments and evaluate billions of system-wide network hypotheses that can best explain the data having a sufficient number of patients or biological samples of appropriate genetic diversity and/or the use of appropriate animal models as surrogates for human diseases. Groups such as ours have developed technology that formalizes these approaches and applies them to the discovery of biomarkers and drug targets. This work led to the discovery of several novel drug targets and drug candidates (E Schadt, personal communication). Bayesian network inference algorithms applied to these well- designed data sets revealed a complex interconnected circuitry of several hundred genes that drive the low density lipoprotein, high density lipoprotein, and triglyceride metabolism, and body mass index of these mice that was subsequently validated in a large human cohort. Here the genetic diversity acted as the ‘ perturbation ’ or ‘ driver ’ that allowed correlation between various components to be interpreted as ‘ causal ’ network interactions. Sieberts and Schadt pioneered the creation and application of methods that linked genetic alterations to transcriptional changes and physiological outcomes in a genetic cross of two inbred mouse strains, one with a propensity for obesity and diabetes, and the other with a genetic resistance to obesity and diabetes. But some exciting progress has been made. We are still far from having comparable blueprints for the pathophysiologies of disease: connections are still miss- ing, and systematic measurements to parameterize and otherwise validate models are still hard to make. Black box models also work, provided the inputs and outputs are robustly validated. Classical engineering solutions depend on a complete and accurate blueprint of the system. While many of these efforts are impressive and in some rare cases have the ability to make non-intuitive predictions supported by experiment, these approaches address only a tiny fraction of the total possible circuitry of human biology and disease. Two contrasting approaches for unraveling the behavior of underlying genetic circuits and biochemical pathways are: (a) the curation of well-known biological pathways from the literature and conversion to descriptive visual displays depicting them as maps to visualize molecular profile changes in their context, Ingenuity Systems, and GeneGo Metacore pathway analysis] and (b) the construction of systems of differential equations that model the time evolution of gene products and their connection to phenotypic changes. a number of different approaches to interpret- ing large sequencing datasets and, increasingly, these fall within the realm of systems or network biology. Your veterinarian may supplement the required vitamins and minerals through medications or suggest an alternative cat kibble. Clinically significant cerebellar edema, usually manifest by a reduced level of consciousness, typically occurs within 1 to 7 days, with a mean peak of 3 days. Iron deficiency or Anemia can cause such in kids and pregnant women. Depending on what is ingested, consumption of these non-food items can lead to intestinal blockages, toxic side effects, or other health issues. It can be there due to a variety of reasons such as. Malnourished felines may chew on inappropriate objects if their diet is lacking in adequate nutrients. Pica is an eating disorder that causes someone to crave and compulsively eat things that are not food, such as paint chips, dirt, paper, or hair. Keeping household plants, blankets, clothing and electrical cords out of your cat’s reach will remove the temptation to eat them.Ĭat toys and safe plants like catnip can detour the feline’s behavior to a more appropriate chewing object.īoredom is a common cause for pica, so structured playtime and exerciser with the feline can prevent boredom and fulfil the need to be active. If the veterinarian has found an underlying disease, the treatment will be specified by the veterinary medical professional, but if your feline has received a clean bill of health, treatment may include: The treatment for pica in cats is variable, as it lies dependent on the underlying cause and the results from diagnostic exams. Pica is a serious behavioral issue that can become fatal if not addressed by a veterinarian. Other inedible objects, such as clothing and plastic, can block the intestine and prevent food from passing. Pica is a condition in which a person craves substances that are not food, such as clay, dirt, ice, or flecks of old paint. Eating non-food items can be very dangerous to a cat, as chewing on electrical cords can cause a feline to be electrocuted, and plant consumption can be toxic. Pica in cats is the act of eating objects that are not food. If your cat licks, sucks, or consumes objects around the home that are not food, she could be suffering from a condition called pica. Oriental cat breeds, such as the Siamese cat, are more commonly affected by pica and there is believed to be a genetic disposition. Signs and Symptoms of Pica Persistent eating of non-nutritive, non-food substances over a period The eating of non-nutritive, non-food substances is. The cause behind pica is unknown, however, disease and behavioral disorders are thought to be the underlying cause behind this unusual behavior. Common target objects for feline pica include: Plants, electric cords, phone cords, wool, fabric, string, or yarn. However, in severe cases of pica, the feline will consume the object entirely, posing a risk for intestinal blockage, tearing of the digestive tract, toxicity, and electrocution. How Is Pica Treated Given the risk of medical complications (such as lead poisoning) associated with pica, close medical monitoring is necessary throughout treatment of the eating behavior. Pica symptoms are caused by swallowed toxic or poisonous substances, as well as bacteria found in nonfood things.A feline with a mild case of pica may suck or lick on inedible objects, but not actually consume said object. Pica is the pathological craving for and eating of a nonnutritive item (eg, clay, coal, paper) or food ingredients (flour, raw potatoes). Pica involving dozens of additional substances has been documented, including cigarette butts and ashes, hair, paint chips, and paper. Eating earth, soil, or clay (geophagia) ice (pagophagia) and starch (starchophagia) are some of the most often documented varieties of pica (amylophagia). There are multiple eponymous posterior stroke syndromes, often presenting incompletely 4. To produce these versions, the software was completely rewritten in C++, while retaining most of the original's functionality and user interface with numerous enhancements. Scores created on one platform could be opened on the other, and were backward compatible. A Mac version 1.2 was released a few months later, and the company thereafter used conventional version numbers for both platforms across subsequent upgrades. In September 1998, the first version for Windows was released as 'Sibelius', with the version number reset to 1.0. 'Lite' versions were subsequently released, and these were successful in UK schools, where Acorns were widely used. It also sold in smaller numbers in a few other countries, restricted by the availability of Acorn computers. Other early adopters included composer John Rutter, conductor Michael Tilson Thomas, and publisher Music Sales.Īs a killer application for the niche Acorn platform, Sibelius rapidly dominated the UK market. The first score published using Sibelius was Antara by George Benjamin, also copied by Emsley, and published by Faber Music. The first concert performance from a Sibelius score was of an orchestral work by David Robert Coleman, copied by Emsley. The first ever user of Sibelius was the composer and engraver Richard Emsley, who provided advice on music engraving prior to the start of development, and beta tested the software before its release. A unique feature of the Sibelius GUI at that time was the ability it gave the user to drag the entire score around with the mouse, offering a bird's eye of the score, as distinct from having to use the QWERTY input keyboard arrow keys, or equivalent, to scroll the page. No matter how long the score, changes were displayed almost instantly. It required considerably less than 1 MB of memory (as its files only occupied a few KB per page of music), and the combination of assembly language and the Archimedes' ARM processor meant that it ran very quickly. The program was released to the public in April 1993 on 3.5-inch floppy disk. Both were music students, and said they wrote the program because they did not like the laborious process of writing music by hand. Development in assembly language on the RISC OS started in 1986 after they left school, and continued while they were at Oxford and Cambridge universities, respectively. Finnish), as well as being one of their favourite composers. The Finns said they could not remember why they used Jean Sibelius' name, but it was probably because he was also ‘a Finn' (i.e. Sibelius was originally developed by British twins Jonathan and Ben Finn for the Acorn Archimedes computer under the name 'Sibelius 7', not as a version number, but reminiscent of Sibelius' Symphony No 7. Avid subsequently recruited some new programmers to continue development of Sibelius, and Steinberg hired most of the former Sibelius team to create a competing software, Dorico. In July 2012, Avid announced plans to divest its consumer businesses, closed the Sibelius London office, and removed the original development team, despite a 12,000 strong 'Save Sibelius' petition spearheading a campaign led by Derek Williams that included extensive protests on Facebook and elsewhere. In August 2006 the company was acquired by Avid, to become part of its Digidesign division, which also manufactures the digital audio workstation Pro Tools. The company won numerous awards, including the Queen's Award for Innovation in 2005. In addition to its head office in Cambridge and subsequently London, Sibelius Software opened offices in the US, Australia and Japan, with distributors and dealers in many other countries worldwide. It went on to develop and distribute various other music software products, particularly for education. Named after the Finnish composer Jean Sibelius, the company was founded in April 1993 by twin brothers Ben and Jonathan Finn to market the eponymous music notation program they had created. Less advanced versions of Sibelius at lower prices have been released, as have various add-ons for the software. It produces printed scores, and can also publish them via the Internet for others to access. Beyond creating, editing and printing music scores, it can also play the music back using sampled or synthesised sounds. It is the world's largest selling music notation program. But she snots through the whole movie, and that’s the key to good acting, right?Īnyway, after Griz squats down and crawls away on all fours (weird), in bounds “Bustopher Jones,” a fat cat in white spats (James Corden - duh). I mean, she’s even sporting a fresh manicure on her blatantly human hand. But a hot Griz kind of defeats the purpose (as if anything in “Cats” has a purpose). It’s as if Grizabella has severe age-dysmorphia, and the cats who make fun of her are just jealous of how fly she is. Nothing about her is “grizzled,” which is particularly apparent when she’s acting alongside Dame Judi Dench and Sir Ian McKellen, who are some truly old-ass cats. When other young actresses played the scraggly, old-ass cat, they were generally aided by ratty costuming and haggard makeup (but which still included a cat-nose - why, oh why, do all these CGI cats have human noses?). Hudson as Grizabella is an interesting choice. I think I’ll chalk this up to an all-white makeup team’s bizarrely misguided attempt at political correctness. Even the iconic shabby grey coat normally worn by Grizabella (played by a hella-dramatic Jennifer Hudson) looks in this version like it may have been designed to match Hudson’s skin tone. His regular fur is brown, just like the fur covering all other cats played by African-American actors. Rum Tum wears a collar (ironically?), and a red fur coat (which is made from, I don’t know, the cat that was sacrificed last year?) over his regular fur. Then “The Rum Tum Tugger” bursts onto the scene a “curious cat” (I’ll say) played by a stilted Jason Derulo, who is clearly plagued by the bitter awareness that he is, in fact, in this movie. I’ve never done LSD, but after watching this Gumby Cat scene, I don’t see a need. Some people-faced beetles trot out next, and they dance and sing with aplomb, but maybe one messed up the choreo or something, because Jennyanydots swiftly grabs and eats it, even as its tiny human face cries out for mercy. So, all Jellicle Cats, even Jellicle Kittens, have adult faces - and all Jellicle Mice (I assume they’re “Jellicle Mice.” I mean, there’s a “Jellicle Moon,” so I’m pretty sure everything in Cat Land is “Jellicle” plus, I’m starting to suspect that “Jellicle” just means “trippy as shit”) look like toddlers? Is Jennyanydots keeping a box of toddlers in her basement? See, these CGI mice - much like these CGI cats - have entirely human faces. Well, once J-Dot opens up a box of mice, the zipper conundrum becomes quite trivial. So, is what we’d assumed to be her fur really just fur-shaped clothing, and is the vest really just clothing-shaped fur? Or does that vest-fur unzip, too? Are Gumby Cats layered, like onions? Can they stack inside one another, like Russian dolls? (If you’re having trouble following, just imagine a naked human suddenly ripping all their skin off to reveal a sparkly tuxedo underneath.) And underneath that fur is (drumroll, please) more fur! But this fur has a vest over it. (Munku doesn’t phrase it quite like that, but that’s my takeaway.) This Gumby Cat (Rebel Wilson) is named “Jennyanydots,” but that’s irrelevant because she’s only ever referred to as “The Gumby Cat.”Īll this is straight from the source. So, after the Jellicles explain to Victoria what a “Jellicle Cat” is (spoiler: they explain nothing), Munkustrap the tour-guide cat whisks Victoria away to observe some sort of Jellicle subspecies called a “Gumby Cat:” a psychotic beast that pretends to be a lazy lump until nightfall, when it races to the cellar and forces its rodent hostages to perform for its pleasure. Eliot’s poems about cats, featuring cats singing about cats - and if I tried to unpack everything that’s cuckoo about the musical itself, this article would never end I’ll try to stick to the ways in which the movie miraculously manages to outweird the musical. Now to be fair, the “plot” of the movie isn’t much of a divergence from the source material - Andrew Lloyd Webber’s operetta about cats, based on T.S. Look for a provider with a pricing plan that gives you enough conference calling time and participants for your conferencing needs. Most pricing plans come with a limited number of participants and cap your calls at 40 minutes. The number of users that you can host in your audio or video conferencing is critical for choosing a conference call service provider. You should be able to easily use essential features like call forwarding, meeting recording, inviting participants, and scheduling calls. Your conference call service should have a user-friendly interface to make it easy to use for all participants, including your staff. The best service providers also offer FAQ pages and informative articles for further reading. In addition to standard support channels such as live chat, email, and phone, look for service providers with live support and the option to contact an agent by speed-dialing. You need responsive, 24/7 customer support available via multiple communication channels. The conference call service provider you choose must provide reliable customer support when you need it the most. Although they may incur a higher price, pricing plans provide extra features that most businesses need. Unlimited monthly or annual pricing plans require a reservation-less service, allowing users to host meetings on demand without limitations. It typically charges the user a per-minute fee for each used functionality. The pay-as-you-go pricing structure allows you to pay only for the features you use. A cost-effective conference call solution should be secure, unlimited, and uninterrupted, with mobile and desktop interfaces, responsive, 24/7 customer support, and full-suite features with exceptional audio and video quality.Ĭonference call service providers usually offer two pricing plan types: pay-as-you-go and unlimited use for a fixed charge. It’s important to look for a service that provides the most value for your money. It allows you to determine the total value you can receive from a service provider in terms of add-on fees, pricing plan costs, and features. Pricing is a vital factor when comparing the best conference call services. In addition to easy set-up of audio and video calls, you also need extras for collaboration and facilitating discussions with advanced reporting and tracking parameters. Opt for a service with a conferencing solution that can help make your conference calls more effective and well-organized. Most users generally choose a provider with uninterrupted and unlimited services with high-quality audio and video. Conference call featuresĪlthough the web offers countless conferencing services, most of them don’t provide the same suite of features and functionalities. The best conference call service provider should provide high-end features, affordable pricing, reliable customer support, a user-friendly interface, exceptional audio/video quality, advanced analytics, and more. There are a couple of other possibilities, but those are the most common ones I encounter. If it doesn't, sometimes making the job for the specific workshop will reset it. The easiest way I've found to fix that is to delete the job from the manager, cancel all jobs out of the workshop and then remake the job in the manager. Sometimes a workshop gets confused and keeps spamming out job orders even though they shouldn't be there. If that's *not* the problem, then it can be other things. Dwarf Fortress building, stockpile, and zone names have a maximum length. It's a pain, but sometimes the 'p' condition is missing and that's the fastest way I know to figure out what it needs to be. These aliases give descriptive names to workshops, levers, stockpiles, zones, etc. See what it is looking for and use *that* for your 'p' condition in the previous step (in this case gathering sand). So in this case, you want to make glass, so make a manager glass job and add an 'r' condition. Once a field is prepared in Dwarf Fortress, you’ll need to choose a crop to plant for each of the four seasons. The easiest way to check what you need to look for is to go backwards in the chain. In this case, I think you will find that it will not change and the reason is that you need "sand-bearing items" (that's from memory, so if it's wrong I apologize - I have no sand on my current embark to check). The Dwarf Fortress Steam/itch release date is December 6th, this year We'll also try to get Classic up here as soon as we can - with a good November I should be able to finish it as well, but I have to get Steam Workshop sorted and I've never tried anything like it before. If it never changes it's because the condition is wrong somehow. Set the number so that the condition is red. My fort is humming along nicely now that I've got an entire surplus furniture stockpile jam packed with pots and bins, in turn jam packed with bags.Īlthough, I suppose that if you really wanted you, you could maybe forbid the seeds and tell your dwarfs to ignore food via the general orders.Have some sand bags in your stockpile (if the jobs are going non-stop, then you probably already have lots). so you can make more thread, and use that thread to make enough bags :p I hope some of those seeds are for pigtails. It sounds like you have a bit of a deficit anyway. More preferably, though, Make enough bags and they'll run out of seeds, and thus subsequently stop putting seeds in them. When there is no longer a place to put the seeds, they'll just drop them. Plants are also where booze comes from, and you do not want to jeopardize the wellspring of your dwarfs' inebriation. Create barrels and bins as soon as posible to compress gems, cloth, and food. These include precise controls over where a stockpile will. A furniture stockpile for barrels nearby is next. You will not be able to use raw clear glass for anything except for cutting it at a jeweler's workshop or b uilding with it. The Dwarf Fortress Steam/itch release date is December 6th, this year Well also try to get Classic up here as soon as we can - with a good November I should be able to finish it as well, but I have to get Steam Workshop sorted and Ive never tried anything like it before. The seeds are important so you will always have enough food growing to not starve. Dwarf Fortress comes with some tools that make running a heavily industrialized fortress easier. Once the clear glass window is crafted it will be carried by a dwarf to a furniture stockpile, at which point you can b uild it. Then specify a tiny stockpile for only seeds. Make sure to remove seeds from all other stockpile allowances via their ->ettings. If it’s too fast or too slow, your Pistons won’t work properly. This is important to keep the machine running at the proper speed. Place a Redstone Repeater, set to two ticks (right click it once after placing) which delays the signal by 1 redstone tick. The comparator pulls the signal from the clock and transfers it into the solid block. Then, place a Redstone Comparator on A2 facing another solid block on A3. Place any solid block on C1 and attach a lever to it to stop the Redstone Clock from ticking. This is what is known as a Redstone Clock, which outputs a signal of one redstone tick per transfer (or approximately 0.1 second intervals). The hoppers should look how they do in the fourth screenshot. Place any object inside and you will notice the object switch between the two hoppers. Then place two hoppers facing each other on A2 and B2. The Sticky piston at D2 will be pushing and pulling the farmland you will use. Step 1: Create a 3x4 area with sticky pistons (A1 to D3).
